A Fast Biomedical Solution to Diagnostic and Academic Uses

Here we will specifically talk about tuberculosis (short as MTB or TB). It was ever called phthisis, phthisis or pulmonalis, is a widespread, and in many cases fatal, infectious disease that typically affects human lungs. The bacteria that cause tuberculosis are spread from one person to another through tiny droplets released into the air via active TB infection cough, sneeze, or otherwise transmit respiratory fluids. For most cases, there are no clinical symptoms, called latent tuberculosis, however 10% of these cases might eventually become active and lead to a 50% death for infected ones. It causes concerns both in developing and developed countries.

Therefore TB prevention and control efforts never stopped. Currently the primarily ways for us to do precaution is to rely on the vaccination of infants and the detection and appropriate treatment of active cases. The first step is to find out if someone is infected if he/she still doesn‘t know. But how?

As a rapid and an effective management method, Tuberculosis rapid tests, also known as one-step TB tests could be a great choice.  
Tuberculosis test kit is a kind of chromatographic immunoassay (CIA) that gives qualitative detection of tuberculosis antibody in human serum or plasma samples. It is designed to be used as a screening test and as an aid in the diagnosis of infection due to Mycobacterium tuberculosis. With the time serum or plasma samples are added to sample window of the TB test cassette, the assay test result will be showed in about 15 minutes. This test is easy to perform and fast to get result.

The test cassette is made of:
1. A burgundy colored conjugate pad that contains TB antigens conjugated with colloid gold and rabbit IgG-gold conjugates;
2. A nitrocellulose membrane strip containing two test bands (T1 and T2 bands) and a control band (C band). The T1 band is pre-coated with monoclonal anti-human IgM for the detection of IgM anti-TB, the T2 band is pre-coated with reagents for the detection of IgG anti-M.TB, and the C band is pre-coated with goat anti-rabbit IgG.

About the results

When an adequate volume of test sample is dispatched into the sample window, the sample migrates by capillary action across the cassette. IgM anti-M.TB if present in the sample will bind to the M.TB conjugates. The immunocomplex is then captured on the membrane by the pre-coated anti-human IgM antibody, forming a burgundy colored T1 band, indicating a M.TB IgM positive test result. IgG anti- M.TB, if present in the sample, will bind to the M.TB conjugates. The immunocomplex is then captured by the pre-coated reagents on the membrane, forming a burgundy colored T2 band, indicating a M.TB IgG positive test result. Absence of any T bands (T1 and T2) suggests a negative result. The test contains an internal control (C band) which should exhibit a burgundy colored band of the immunocomplex of goat anti rabbit IgG/rabbit IgG-gold conjugate regardless of the color development on any of the T bands. Otherwise, the test result is invalid and the sample must be retested with another device.
